不同雷公藤制劑中6個成分的含量研究

摘 要:目的 建立測定雷公藤制劑中的6種成分(雷公藤甲素、雷公藤內酯酮、雷酚內酯、雷公藤次堿、雷公藤紅素及雷公藤內酯甲)的高效液相色譜方法。方法 采用Agilent TC-C18柱(250 mm×4.6 mm,5 μm),以10 mmol/L乙酸銨水溶液-乙腈為流動相梯度洗脫,流速為0.75 ml/min,檢測波長為225 nm,柱溫為25 ℃。結果 6種成分的進樣濃度為0.1~156.25 μg/ml范圍內與峰面積積分值線性關系良好,平均回收率為98.5%(RSD=1.9%)、99.2%(RSD=1.9%)、99.5%(RSD=1.5%)、98.7%(RSD=1.2%)、98.8%(RSD=1.6%)、99.0%(RSD=1.7%)。結論 所建立的測定方法操作方便,重復性強,結果準確,可用于雷公藤及其相關產品的質量分析;不同雷公藤制劑中6個成分的含量存在差異。

Abstract:Objective To establish a high performance liquid chromatography method for the determination of 6 components in Tripterygium wilfordii(Triptolide,triptolide,lycopolide,Tripterygium,tripterin,and triptolide a).Methods A Agilent TC-C18 column of (250 mm×4.6 mm,5 μm)was used.The mobile phase gradient elution was 10 mmol/L ammonium acetate aqueous solution and acetonitrile.The flow rate was 0.75 ml/min,the detection wavelength was 225 nm,and the column temperature was 25 ℃.Results The injection concentration of the six components was 0.1~156.25 μg/ml and the peak area integral value was linear,the average recovery rate was 98.5%(RSD=1.9%),99.2%(RSD=1.9%),99.5%(RSD=1.5%),98.7%(RSD=1.2%),98.8%(RSD=1.6%),99.0%(RSD=1.7%).Conclusion The established method is convenient, reproducible and accurate.It can be used for the quality analysis of Tripterygium wilfordii and its related products,and the content of 6 components in different Tripterygium wilfordii preparations is different.
