黔產(chǎn)苗藥對(duì)坐葉藥材HPLC指紋圖譜研究

摘 要 目的:建立黔產(chǎn)苗藥對(duì)坐葉藥材的高效液相色譜(HPLC)指紋圖譜。方法:采用HPLC法。色譜柱為Agilent Eclipse XDB C18,流動(dòng)相為乙腈-0.2%磷酸溶液(梯度洗脫),流速為1.0 mL/min,檢測(cè)波長(zhǎng)為235 nm,柱溫為25 ℃,進(jìn)樣量為15 ?L。以蘆丁為參照,測(cè)定10批貴州不同地區(qū)對(duì)坐葉藥材的HPLC圖譜,采用《中藥色譜指紋圖譜相似度評(píng)價(jià)系統(tǒng)》(2004A版)進(jìn)行共有峰指認(rèn)和相似度評(píng)價(jià)。結(jié)果:10批對(duì)坐葉藥材的HPLC指紋圖譜有12個(gè)共有峰,相似度均大于0.90。經(jīng)驗(yàn)證,10批樣品HPLC圖譜與對(duì)照指紋圖譜具有較好的一致性。結(jié)論:本研究所建HPLC指紋圖譜可為對(duì)坐葉藥材的鑒別和質(zhì)量評(píng)價(jià)提供參考。

ABSTRACT OBJECTIVE: To establish HPLC fingerprint of Miao medicine Hedyotis uncinella from Guizhou. METHODS: HPLC method was adopted. The determination was performed on Agilent Eclipse XDB C18 column with mobile phase consisted of acetonitrile-0.2% phosphoric acid (gradient elution) at the flow rate of 1.0 mL/min. The detection wavelength was set at 235 nm, and column temperature was 25 ℃. The sample size was 15 ?L. Using rutin as reference, HPLC chromatograms of 10 batches of H. uncinella from different areas of Guizhou province were determined. Similarity Evaluation System for TCM Chromatographic Fingerprints (2004A edition) was used to identify common peaks and evaluate similarity. RESULTS: There were 12 common peaks in HPLC fingerprints of 10 batches of H. uncinella, and the similarity was higher than 0.90. After validation, HPLC chromatograms of 10 batches of sample were in agreement with control fingerprints. CONCLUSIONS: Established HPLC fingerprints can provide reference for the identification and quality evaluation of H. uncinella.

苗藥對(duì)坐葉為茜草科植物對(duì)坐葉(Hedyotis uncinella Hook. et Arn.)的干燥全草,為貴州苗藥。對(duì)坐葉味辛、微苦,性平,具有清熱解毒、調(diào)中止瀉的功效,可用于治療腹瀉、腹脹、腹痛、里急后重以及痢疾、急性胃炎。苗藥解毒止瀉膠囊,由苗藥對(duì)坐葉為主藥組成中藥復(fù)方制劑。目前該產(chǎn)品主要在遵義家誠(chéng)藥業(yè)生產(chǎn),苗藥解毒止瀉膠囊用于胃腸濕熱所致的腹瀉、腹脹、腹痛、急性腸炎等癥狀,作為地方藥材標(biāo)準(zhǔn)被收載于《貴州省中藥材、民族藥材質(zhì)量標(biāo)準(zhǔn)》(2003年版)[1],F(xiàn)行標(biāo)準(zhǔn)僅通過(guò)性狀、顯微、薄層對(duì)對(duì)坐葉藥材進(jìn)行質(zhì)量控制。本文對(duì)對(duì)坐葉藥材進(jìn)行高效液相色譜(HPLC)指紋圖譜研究,并對(duì)10批不同產(chǎn)地來(lái)源對(duì)坐葉藥材進(jìn)行了相似度評(píng)價(jià)比較,建立HPLC指紋圖共有模式,以期為對(duì)坐葉藥材的質(zhì)量控制和有效識(shí)別提供參考。
1 材料
1.1 儀器
LC-1260型HPLC儀,包括四元泵、二極管陣列檢測(cè)器、柱溫箱、自動(dòng)進(jìn)樣器、Agilent 1260工作站(美國(guó)Agilent 公司);AB204-S型電子分析天平(瑞士Mettler-Toledo公司);KQ-500DV型數(shù)控超聲波清洗機(jī)(功率:500 W,頻率:60 kHz,昆山市超聲儀器有限公司);111型中藥粉碎機(jī)(上海海久中藥機(jī)械制造有限公司)。
1.2 藥材、藥品與試劑
對(duì)坐葉藥材采集于貴州各地區(qū),經(jīng)遵義醫(yī)藥高等專科學(xué)校劉亮副教授鑒定為茜草科植物對(duì)坐葉(Hedyotis uncinella Hook.et Arn.)的干燥全草。藥材來(lái)源及信息見(jiàn)表1。蘆丁對(duì)照品(批號(hào):100080-200707,純度:100%,購(gòu)于中國(guó)食品藥品檢定研究院);乙腈為色譜純,其余試劑均為分析純,水為純水。
2 方法與結(jié)果
2.1 色譜條件
色譜柱:Agilent Eclipse XDB C18(250 mm×4.6 mm,5 μm);流動(dòng)相:乙腈(A)-0.2%磷酸溶液(B),梯度洗脫(0~5 min,5%A→10%A;5~15 min,10%A→15%A;15~25 min,15%A→20%A;25~35 min,20%A→30%A;35~40 min,30%A→5%A;40~45 min,5%A);流速:1.0 mL/min;柱溫:25 ℃;檢測(cè)波長(zhǎng):235 nm;進(jìn)樣量為15 μL;運(yùn)行時(shí)間:45 min[2-6]。
