HPLC—ELSD法測定不同儲存時間黨參藥材中果糖、葡萄糖和蔗糖的含量

[摘要] 目的 建立黨參藥材中果糖、葡萄糖和蔗糖高效液相色譜-蒸發(fā)光散射檢測法(HPLC-ELSD)含量測定的方法,并測定不同儲存時間的黨參果糖、葡萄糖和蔗糖的含量。方法 采用80%甲醇超聲提取,通過HPLC-ELSD法測定不同儲存時間的黨參中果糖、葡萄糖和蔗糖的含量。色譜條件:Shodex Asahipak NH2P-50 4E(250 mm × 4.6 mm,5 μm)色譜柱,以乙腈-水(77∶23)為流動相,柱溫25 ℃,流速1.0 mL/min,ELSD檢測器。結(jié)果 通過實(shí)驗(yàn),不同儲存時間的黨參中果糖、葡萄糖和蔗糖均能很好分離,線性范圍和相關(guān)系數(shù)分別為2.409~24.09 μg(r = 0.9999)、1.456~14.56 μg(r = 0.9995)、1.15~11.50 μg(r = 0.9999),平均回收率分別為103.73%(RSD = 0.26%)、97.85%(RSD = 1.98%)、99.08%(RSD = 1.09%)。結(jié)論 表明該方法簡便,穩(wěn)定性、重復(fù)性、分離度好,可用于黨參中果糖、葡萄糖和蔗糖含量分析,儲存0、3、6、12個月的不同時間黨參中果糖、葡萄糖和蔗糖含量差異明顯。

[Abstract] Objective To establish a HPLC-ELSD determination method of fructose, glucose and sucrose in Codonopsis pilosula, and to determine the content of fructose, glucose and sucrose of Codonopsis pilosula in different storage times. Methods The sample solutions made by ultrasonic extraction of 80% methanol solution were used to determine the contents of fructose, glucose and sucrose of Codonopsis pilosula in different storage times by HPLC-ELSD. The analysis was performed on a Shodex Asahipak NH2P-50 4E column (250 mm × 4.6 mm, 5 μm) with a gradient elution of acetonitrile-water (77∶23) at the flow rate of 1.0 mL/min. The column temperature was 25℃. It was determined by ELSD detector. Results The methodological studies showed that fructose, glucose and sucrose of Codonopsis pilosula in different storage times were in good linearity from 2.409 to 24.09 μg (r = 0.9999), 1.456 to 14.56 μg(r = 0.9995), 1.15 to 11.50 μg (r = 0.9999) respectively. The average recoveries of the three components were 103.73% (RSD = 0.26%), 97.85% (RSD = 1.98%) and 99.08% (RSD = 1.09%), respectively. Conclusion The results show that the method is convenient, stable, reproducible and well separated. It can be used to determine fructose, glucose and sucrose in Codonopsis pilosula. The content of fructose, glucose and sucrose in Codonopsis pilosula of different storage times(0, 3, 6, 12 months) are different obviously.

黨參來源于桔梗科植物黨參Codonopsis Pilosula (Franch.)Nannf.、素花黨參Codonopsis pilosula Nannf. var. modesta(Nannf.)L. T. Shen 或川黨參Codonopsis tangshen Oliv. 的干燥根[1]。黨參根據(jù)產(chǎn)地分為西黨參、東黨參和潞黨參等[2]。黨參所含主要化學(xué)成分有苷類、糖類、甾醇、膽堿、生物堿、木質(zhì)素、三萜等[3-9]。文獻(xiàn)報道[10-16],黨參糖類成分為黨參具有滋補(bǔ)功效的有效成分之一,具有調(diào)節(jié)血液系統(tǒng)、抗癌、抗炎、抗病毒、抗衰老、抗細(xì)胞毒性等免疫增強(qiáng)的功效。由于黨參藥材中果糖、葡萄糖、蔗糖含量很高,在儲存過程中容易發(fā)生糖的轉(zhuǎn)化與降解以及容易發(fā)生霉變等過程中造成糖的損失。有文獻(xiàn)報道了黨參不同儲存時間的質(zhì)量考察研究,但該研究只考察了氣味、顏色、霉變等指標(biāo),沒有考察指標(biāo)性成分和糖類成分的含量變化[17]!吨袊幍洹2015年版黨參項(xiàng)下未建立含量測定項(xiàng),也未見有文獻(xiàn)報道同時測定黨參藥材中果糖、葡萄糖、蔗糖的含量。本文首次建立了HPLC-ELSD法測定果糖、葡萄糖、蔗糖含量的一針同時測定3個指標(biāo)成分的方法,可用于黨參藥材的質(zhì)量評價,研究的不同儲存時間黨參中果糖、葡萄糖、蔗糖含量差異明顯,為黨參藥材的儲存期提數(shù)據(jù)支持。
