三七皂苷對硫化氫致豚鼠急性肺損傷的治病機制的研究

[摘要] 目的 觀察三七皂苷(SAT)對豚鼠肺損傷的治療作用并研究其作用機制。 方法 120只豚鼠隨機分為T0組、TS組、SAT組、FS組。采用硫化氫制備豚鼠肺損傷模型,構模8 h后停止給藥,乙醚麻醉動物,分別于7 h、14 h、21 h及24 h時間點觀測其肺損傷程度及病理變化。 結果 在各時間點觀察:SAT能有效降低SWW濃度及肺靜脈高壓,與T0組、TS組比較差異具有統(tǒng)計學意義(P<0.05);但SAT與激素聯(lián)合治療效果差異并不明顯,與FS組比較差異無統(tǒng)計學意義(P>0.05)。 結論 SAT能有效抑制硫化氫引起的肺損傷進程。

Mechanism of panax notoginseng saponins on acute lung injury caused by hydrogen sulfide in guinea pigs

[Abstract] Objective To investigate the effective and mechanism of panax notoginseng saponins(SAT) on acute lung injury(ALI) caused by hydrogen sulfide in guinea pigs. Methods 120 guinea pigs were randomized into group T0, group TS, group SAT and group FS. The ALI model of guinea pigs was established by hydrogen sulfide. The drug was removed 8 hours after the model was established. The animals were anaesthetized by aether. The degree of lung injury and pathological changes were observed at 7th, 14th, 21st and 24th hour. Results Compared with group T0 and TS, SAT could decrease the level of SWW and high pulmonary venous pressure significantly at every time point(P<0.05). However, compared with group FS, the effectiveness of SAT was not significantly different from that of combined treatment of hormone(P>0.05). Conclusion SAT could inhibit the process of lung injury caused by hydrogen sulfide effectively.

三七(Pseudo-ginseng)作為一種重要中藥材,得到臨床廣泛的應用,對其藥理研究也被現(xiàn)在的中外學者不斷論述,但是多都停留在分子前水平,通常為三七總皂苷(SAT)[1]。本實驗即在此基礎上探尋SAT對阻止一些急性損傷因素中所起的關鍵作用[2]。肺水腫常是某些損傷性疾病的前期癥狀,盡管病因多種,但病理基礎基本相同[3],實驗中以硫化氫作為致模藥物,選擇豚鼠構造肺水腫模型[4],實驗前經(jīng)過反復測試得出標準構模時間、標本重量、藥物濃度等重要參數(shù),實驗證實三七皂苷應用于急性肺損傷治療,能夠有效降低肺靜脈高壓及SWW濃度,阻止急性肺水腫發(fā)展,為臨床應用提供重要依據(jù)。
1材料與方法
1.1實驗材料
1.1.1動物 健康雄性豚鼠:沈陽醫(yī)學院動物模型中心Sy1703-S003組120只,體重310~350 g,平均340 g。
1.1.2構模材料 硫化氫(沈陽景泉特種氣體廠,H2S,Q1000~07號)。
1.1.3實驗藥品、儀器、設備 見表1。
1.2 實驗方法
、賱游锬P偷臉嫿ǎ毫蚧瘹洌50 mL/L)密室200 m3,將豚鼠全部投放密室經(jīng)8 h取出,進行乙醚罩吸麻醉,1.5%戊巴比妥鈉注射液靜脈加強麻醉。②分組:將120只構型豚鼠隨機分成四組,每組30只,置于實驗臺連接實驗設備、儀器。Ⅰ組對照組(T0組):100 mL生理鹽水肺靜脈連續(xù)滴注;Ⅱ組(TS組):5 mg地塞米松加生理鹽水100 mL肺靜脈連續(xù)滴注;Ⅲ組(SAT組):生理鹽水100 mL加三七皂苷0.25 g配制成濃度0.25%肺靜脈連續(xù)滴注;Ⅳ組(FS組):在SAT組基礎上聯(lián)合地塞米松5 mg肺靜脈連續(xù)滴注,測量肺毛細血管靜脈壓(PCWP)值,記錄肺毛細血管靜脈壓值、血氧飽和度(FIO2)值,各組于造模后7 h、14 h、21 h、24 h觀察記錄實驗數(shù)據(jù)。
