摘 要：再造煙葉總植物堿測定采用YC/T 468-2013的方法，但是在測定時會遇到緩沖溶液B很難溶解的問題，即使加熱超聲溶解后，也會出現(xiàn)結(jié)晶析出，容易導致管道堵塞。為了解決此問題，通過減少緩沖溶液B中磷酸氫二鈉的用量的方法測定再造煙葉總植物堿含量，并將測定結(jié)果與標準方法測定結(jié)果進行對比分析。結(jié)果表明：緩沖溶液B磷酸氫二鈉用量為130 g時，對再造煙葉總植物堿測定結(jié)果沒有影響；改進后的檢測方法檢出限和定量限分別為0.057 2%和 0.202 8%，相對標準偏差為（0.19～0.55）%。改進后的方法適合再造煙葉總植物堿含量的測定。

　　An Improved Method for Determining Total Alkaloid
　　Content in Reconstituted Tobacco

　　Abstract： The YC/T 468-2013 method is usually used to determine total alkaloid content in reconstituted tobacco， but it is difficult to dissolve the buffer solution B during determining. Its crystal precipitation can also appear after dissolving by heating and ultrasonic， which will cause the clogging in the pipe. In order to solve the problem， the amount of disodium hydrogen phosphate in the buffer solution B was decreased， and determined results of the total alkaloid content in reconstituted tobacco were compared with analyzed results. The results show that： when the dosage of disodium hydrogen phosphate in buffer solution B is 130 g， it has no effect on determined result of total plant alkaloid content； the detection limit and the quantitative limit of improved detection methods respectively are 0.0572% and 0.202 8%， and relative standard deviation is 0.19%～0.55%.The improved method is suitable for determining the total alkaloid content in reconstituted tobacco.

　　再造煙葉又名重組煙葉或煙草薄片，已廣泛用作卷煙原料[1，2]，對改善和提高卷煙產(chǎn)品的質(zhì)量，降低卷煙中焦油等有害物質(zhì)的產(chǎn)生量等方面，均具有十分顯著的效果。煙草生物堿以煙堿最為重要，它占煙草生物堿含量的95%以上[3]，其次是去甲基煙堿、新煙堿等，故測定煙草總植物堿的含量時，常以煙堿計。過高的煙堿含量會嚴重影響煙葉品質(zhì)，降低煙葉在卷煙配方中的可用性[4]。通過檢測煙堿含量，可以在生產(chǎn)中降低煙葉煙堿含量，提高煙葉的可用性。目前，我國測定煙草及煙草制品中總植物堿的標準方法有三個[5-7]，分別是GB/T 23225-2008《煙草及煙草制品 總植物堿的測定 連續(xù)流動法》、YC/T 160-2002《煙草及煙草制品 總植物堿的測定 連續(xù)流動法》和YC/T 468-2013 《煙草及煙草制品 總植物堿的測定 連續(xù)流動（硫氰酸鉀）法》。本文主要是針對YC/T 468-2013中規(guī)定的方法進行改進的。
　　1 反應原理[8]
　　在水溶液中二氯異氰尿酸鈉將發(fā)生二級水解，此過程將釋放出ClO-，硫氰酸鉀絕大部分將電離成SCN-。兩種水解的產(chǎn)物ClO-和SCN-瞬間發(fā)生氧化還原反應產(chǎn)生目標化合物ClCN。再造煙葉萃取液中的總植物堿（以煙堿計）與對氨基苯磺酸和氯化氰反應，反應產(chǎn)物用比色計在460 nm測定。
　　2 存在的問題
　　在測定實驗中，YC/T 468-2013中規(guī)定的緩沖溶液B的配置方法為：稱取222 g 磷酸氫二鈉、8.4 g檸檬酸、7.0 g對氨基苯磺酸至燒杯中，用水溶解，然后轉(zhuǎn)入1 000 mL 容量瓶中，用水定容至刻度[7]。磷酸氫二鈉的溶解性為218 g/L（20 ℃），在（11-2）月溫度較低的期間，緩沖溶液B很難溶解，即使加熱超聲溶解后，也會出現(xiàn)結(jié)晶析出。這就直接導致了配置溶解緩沖溶液B的時間過長，大致需要50 min，檢測總植物堿時，緩沖溶液B易析出晶體，導致管道容易堵塞。
